چکیده

As most intracellular signaling takes place via cascades of phosphorylation and dephosphorylation of tyrosines, protein tyrosine phosphatases have emerged as new and promising targets. Among them, protein tyrosine phosphatase 1B (PTP1B) negatively regulates insulin signaling by dephosphorylation of key tyrosine residues within the regulatory domain of the β-subunit of the insulin receptor, thereby attenuating receptor tyrosine kinase activity. Echelby et al. have demonstrated that PTP-1B knock-out mice fed with high-fat diet showed enhanced insulin sensitivity without any adverse symptoms. Thus, PTP-1B inhibitor could potentially ameliorate insulin resistance and normalize plasma glucose and insulin without inducing hypoglycemia. Recently, small molecule inhibitors of PTP-1B as well as peptide mimetics were reported in literatures. They included catechol A, o-quinones B-D, and carboxylic acids E-G. One of the inhibitors, Ertiprotafib (G) went to clinical trial, but was discontinued in Phase II due to insufficient efficacy and dose-dependent side effects. Some catechol derivatives were discovered as hits from high-throughput screening of the library of Korea Chemical Bank. As phenolic compounds with anti-oxidant activities are known to be beneficial in the treatment of diabetes and related disorders, and catechol A and similar compounds

Pyrrolo[2,3-c]azepine derivatives: a new class of potent protein tyrosine phosphatase 1B inhibitors.

A series of pyrrolo[2,3-c]azepine derivatives was designed, synthesized, and evaluated as a new class of inhibitors against protein tyrosine phosphatase 1B (PTP1B) in vitro. The results demonstrated that compounds bearing a biphenyl moiety were proved to markedly influence the potency of these inhibitors. Particularly, compounds 29, 35 and 36 showed interesting inhibition with IC(50) value of 1...

Phosphonate monoesters on a thiacalix[4]arene framework as potential inhibitors of protein tyrosine phosphatase 1B.

Monoester derivatives of thiacalix[4]arene tetrakis(methylphosphonic) acid were found to be capable of inhibiting protein tyrosine phosphatase 1B. In addition, these compounds can strongly bind to human serum albumin.

Cellular inhibition of protein tyrosine phosphatase 1B by uncharged thioxothiazolidinone derivatives.

As important regulators of cellular signal transduction, members of the protein tyrosine phosphatase (PTP) family are considered to be promising drug targets. However, to date, the most effective in vitro PTP inhibitors have tended to be highly charged, thus limiting cellular permeability. Here, we have identified an uncharged thioxothiazolidinone derivative (compound 1), as a competitive inhib...

Penostatin derivatives, a novel kind of protein phosphatase 1b inhibitors isolated from solid cultures of the entomogenous fungus Isaria tenuipes.

Protein tyrosine phosphatase 1B (PTP1B) is implicated as a negative regulator of insulin receptor (IR) signaling and a potential drug target for the treatment of type II diabetes and other associated metabolic syndromes. Therefore, small molecular inhibitors of PTP1B can be considered as an attractive approach for the design of new therapeutic agents of type II diabetes diseases. In a continuin...

The protein tyrosine phosphatase 1b as a drug target for the treatment of diabetes type II. Developing effective and selective PTP1B inhibitors

PTP1B is a protein tyrosine phosphatase involved in insulin receptor desensitization. PTP1B inhibition, resulting in prolonged maintenance of the activated state of the receptor, practically enhances insulin effect. Thus PTP1B has become a drug target for the treatment of Diabetes type II. Although, a great number of inhibitors have been developed, among which inhibitors binding to the catalyti...
